When your computer restarts, it will boot from the USB flash drive. (In many BIOSes, a special key-often the F10 key-will save, exit, and reboot in one step.). Restart/turn on PC and press BIOS key ('F2'/'Esc'/'DEL') to get into 'Profiles Setup Utility'. Plugin the USB flash drive with your Windows framework. After that select the 'F10' key and save all the changes. When you're done with the BIOS setup program, using the tab and/or arrow keys to navigate to the EXIT menu, select the option for saving the changes you just made, and then reboot. But yes, a UEFI system can boot in UEFI mode from either GPT or MBR USB devices as long as the partition is formatted FAT32 and the UEFI bootloader file is there.
